JASCO P-4000 polarimeters

The three-member JASCO P-4000 polarimeter family has replaced the successful P-2000 series after almost two decades. Combining state-of-the-art technology with a space-saving design, the P-4000 polarimeters are designed for high-precision optical rotation measurements. The perfect interaction between hardware and software ensures that the instruments not only meet the requirements of chemical and research laboratories, but also the strict pharmacopoeia and data integrity regulations of pharmaceutical companies.

The standard sample holder of each P-4000 polarimeter is an air-cooled Peltier thermostat, which is suitable for holding 1-10 cm long cuvettes. The temperature can be controlled between 15-40 oC. The temperature sensor that can be placed in the sample can also be used to directly measure and document the sample temperature. In addition to the usual cylindrical polarimeter cells, JASCO’s newly developed RQC square, small volume and easy-to-fill cuvettes with good heat transfer can also be used. The RQC cuvettes are available in 2 cm, 5 cm and 10 cm light path lengths.

The polarimeters can be controlled with a built-in, Windows-based touch screen control unit. The JASCO Spectra Manager-based software provides a neat interface for setting the measurement parameters, viewing and evaluating the measurement results. The measured data can be displayed in the form of rotation (α), specific rotation ([α]), Brix, sugar, concentration and optical purity. The evaluation is assisted by statistical functions calculating the average and standard deviation as well as by the possibility of using temperature and light source correction factors. The results can also be saved onto an external drive in a text-based file format that can be read by other programs as well. In addition to the self-diagnosis function, the instrument status is checked by the daily check and the validation module. During the daily check, information about the operating status of the polarimeter can be obtained by measuring the lamp energy and a standard with known optical rotation. The validation module provides help with periodic checks and includes methods according to the American (USP), European (EP) and Japanese (JP) pharmacopoeias. The basic software of the polarimeters also provides the two-level user distinction and a detailed instrument log (audit trail). To meet the pharmaceutical and other sensitive laboratory requirements in terms of data security, the control can be done with a software that can be installed onto an internal or external computer that meets the requirements of 21 CFR Part 1

The smallest P-4000 polarimeter is the P-4100, which has a long-life LED light source and uses a sensitive photomultiplier (PMT) for detection. The instrument, which is only 44 cm wide, allows measurements with an accuracy of ±0.01 degree.

The P-4200 polarimeter contains the classic sodium lamp as a light source, which allows high-precision measurements on the sodium D-line. The instrument can accommodate 2 light sources at the same time, so it can be supplemented with additional mercury, halogen or LED sources and the appropriate manually replaceable filters.

The optical system of the P-4300 includes both a sodium and a mercury lamp, and filters corresponding to the selected wavelength are inserted into the light path by an automatic filter changer. The 589, 546, 436, 405, 365 nm filters are standard accessories of the instrument. Two light sources can be used simultaneously in the lamp compartment, but the easy exchangeability allows for the optional use of a halogen or LED lamp. In this case, a 633 nm filter can also be used. A Glan-Taylor polarizer and a photomultiplier (PMT) detector ensure the accuracy of the measurements.
